Phenomenex has announced a Chiral Screening Service for customers in pharmaceutical and natural products research and development. This free service provides target screening using a library of HPLC and SFC columns including the Phenomenex Lux polysaccharide-based offerings, which demonstrate a success rate approaching 90 percent.
Twelve chiral derivatizing reagents (CDRs) were synthesized by substituting one of the fluorine atoms in 1,5-difluoro-2,4-dinitrobenzene (DFDNB) with three optically pure amines [(R)-(-)-1-cyclohexylethylamine, (+)-dehydroabietylamine and (S)-(-)- ,4-dimethylbenzylamine], six amino acid amides [L-Ala-NH2, L-Phe-NH2, L-Val-NH2, L-Leu-NH2, L-Met-NH2 and D-Phg-NH2] and three amino acids [L-Ala, L-Val and L-Leu]. The work was led by Professor Ravi Bhushan from the Department of Chemistry at the Indian Institute of Technology Roorkee in India and documented in Biomedical Chromatography [23 (12) 1291 – 1299 (2009)].
In the biotechnology industry, ion-exchange chromatography is widely used for profiling the charge heterogeneity of proteins, including monoclonal antibodies. In a recent paper in Analytical Chemistry [81 (21) 8846–8857 (2009)], a pH gradient based separation using a cation exchange column is described and shown to be a multiproduct charge sensitive separation method for monoclonal antibodies.
A three-parameter expression, previously employed for prediction of gradient mode retention time in reversed-phase high-performance liquid chromatography (RP-HPLC) with satisfactory results, was tested under a variety of gradient patterns, using methanol and acetonitrile as the organic modifiers and documented in Analytical Chemistry [81 (21) 9135–9145 (2009)].
